Efficient Transfection via an Unexpected Mechanism by Near Neutral Polypiperazines with Tailored Response to Endosomal pH

Front Cover: Polypiperazines are hydrophilic pH-sensitive polymers reaching high transfection efficiency and guaranteeing good biocompatibility. They almost charge-neutral under physiological conditions avoiding unintended side effects, while the isopropyl-modified polypiperazine outperforms commercial linear PEI at low N/P ratios. qPCR revealed that pDNA can still be amplified complexed with piperazine-based polymers, providing a promising platform for effective polymeric gene delivery systems. This is reported by Anja Traeger, Johannes C. Brendel, co-workers in article number 2200517.
